摘 要:目的 研究心得安外涂所致豚鼠銀屑病樣動物模型皮損中角蛋白K14,16,17表達(dá)水平的變化,從分子水平研究此種動物模型的可行性. 方法 用50g L-1 心得安乳劑外涂豚鼠耳背部皮膚誘導(dǎo)銀屑病樣動物模型,應(yīng)用原位雜交法檢測銀屑病樣皮損中角蛋白K14,16,17的表達(dá). 結(jié)果 心得安外涂所致豚鼠銀屑病樣動物模型皮損中角蛋白K14,16表達(dá)水平較正常對照組明顯增高,K17的水平無明顯變化. 結(jié)論 心得安外涂所致豚鼠銀屑病樣動物模型不僅在組織病理學(xué)而且在分子水平上與人類銀屑病皮損基本一致.

Abstract:AIM To investigate the changes of keratin14,16,17expression in lesions of guinea pig's psoriasis-like ani-mal model caused by topical treatment with propranolol and explore the feasibility of this animal model at molecular level.METHODS Psoriasis-like animal model was established by topical treatment with5%propranolol emulsion on ear's back of guinea pig.Expression of keratin14,16,17in lesions were detected by in situ hybridization.RESULTS Expres-sion of keratin14,16in lesions of treated group were signifi-cantly increased,while expression of K17was not significant-ly different with that in control group.CONCLUSION Guinea pig's psoriasis-like animal model caused by topical treatment with propranolol has high concordance with human psoriatic lesions not only in histopathology,but at molecular level.

銀屑病的發(fā)病機(jī)制極其復(fù)雜,其組織病理學(xué)的改變主要表現(xiàn)為角質(zhì)形成細(xì)胞過度增殖、炎癥細(xì)胞聚集和真皮乳頭部血管擴(kuò)張三大特征.其中,角質(zhì)形成細(xì)胞過度增殖常常伴有異常的角化過程,同時出現(xiàn)角蛋白表達(dá)的改變.為了能夠更好地研究銀屑病的發(fā)病機(jī)制,合適而科學(xué)的動物模型顯得極為重要.我們從分子水平研究心得安外涂所致豚鼠銀屑病樣動物模型的可行性.
